Antibiotic Screening

  • Antibiotic screening can be used to identify the efficiency of transformation.
  • Reason: Insertion of plasmid vector that containing antibiotic-resistant gene will protect the host cell from being destroyed by antibiotics.
  • Result:​
    • Viable cells (transformant)

      • Host cell with inserted recombinant vector

      • Host cell with inserted normal vector

    • Dead cells (Non-transformant)

      • Host cell without inserted vector
         
         

Blue- white screening

  • Blue-white screening can be used to identify the efficiency of recombination.

  • Reason: Insertion of DNA interest into plasmid vector causing the cell unable to cleave X-gal and appear as white.

  • Result:

    • Blue colonies (Non- recombinant vector)

      •  Vector without inserted DNA

    • White colonies (Recombinant vector)

      • Vector with inserted DNA
