GENE CLONING
Screening
Antibiotic Screening
-
Antibiotic screening can be used to identify the efficiency of transformation.
-
Reason: Insertion of plasmid vector that containing antibiotic-resistant gene will protect the host cell from being destroyed by antibiotics.
-
Result:
-
Viable cells (transformant)
-
Host cell with inserted recombinant vector
-
Host cell with inserted normal vector
-
-
Dead cells (Non-transformant)
-
Host cell without inserted vector
-
-
Blue- white screening
-
Blue-white screening can be used to identify the efficiency of recombination.
-
Reason: Insertion of DNA interest into plasmid vector causing the cell unable to cleave X-gal and appear as white.
-
Result:
-
Blue colonies (Non- recombinant vector)
-
Vector without inserted DNA
-
-
White colonies (Recombinant vector)
-
Vector with inserted DNA
